I remember when Marvel would bill Spidey as the 'Non-mutant superhero', but technically isn't a mutation any organism that's been altered in some way?

Mutation is a change in the genes from one generation to the next. Spidey's 'accident' was in his youth. He was a normal teenager before. So you must name him changed.

If you assume he is 50% Spider and 50% human, and he has children with a human, then theoretically, won't his kids only be 25% spider??
